Development and Characterization of Anti-Morphine Antibodies Through Bovine Serum Albumin Conjugation

Abstract
To produce anti-morphine antibody, first morphine is synthesized with anhydrosuccinic to morphine-6-succinate, and then morphine-6-succinate is conjugated with bovine serum albumin, morphine-6-succinate of bovine serum albumin is obtained. In this research, 8 molecules of morphine succinate were attached to each molecule of bovine serum albumin; Then, morphine-6-succinate of bovine serum albumin was prepared with phosphate buffer of morphine-6-succinate of bovine serum albumin. Then, a dose of immunogen was injected intramuscularly (on days 0, 15 and 30) to adult male rabbits. Of course, before each injection, blood was drawn from the rabbits. On the 45th day, blood was drawn from the rabbits. The method of investigating the presence of anti-morphine antibody activity in the serum of rabbits was enzyme immunoassay. After ensuring the production of antimorphine antibody on days 15, 30 and 45, affinity chromatography method was used to purify the antimorphine antibody. After purification, it was found that the purified antibody was 25 times more It shows activity not purified from serum.
